    Emidio di Treviri, una ricerca sulla gestione del post sisma nel centro Italia: considerazioni sul caos abitativo e le relative ricadute territoriali

    Con questo contributo intendiamo presentare alcuni dei primi risultati di Emidio di Treviri, gruppo di ricerca sul post sisma del centro Italia formatosi nel dicembre del 2016 a seguito di una call delle Brigate di solidarietà attiva, associazione di assistenza mutualistica presente sul cratere del terremoto da agosto 2016. Il nostro contributo, adottando una prospettiva territorialista, si propone di restituire una lettura critica delle diverse soluzioni abitative messe in campo nella fase emergenziale (Cas, strutture alberghiere, ecc.). Tenendo conto delle peculiarità sociali e morfologiche del territorio in cui sono stati proposti, la valutazione sintetica degli strumenti di gestione del post sima che qui presentiamo vuole indagare le ricadute territoriali del caos abitativo generatosi e presentare il quadro delle diseguaglianze e delle vulnerabilità che ne è conseguitoThis paper presents some of the first results of the work of "Emidio di Treviri", a research group which intends to investigate the post-earthquake of Central Italy. The group took shape in December 2016, following a call promoted by the Brigate di Solidarietà Attiva, a mutual assistance association which has been working in the cratere (the earthquake’s damaged area) since September 2016. Our contribution aims to give a critical reading of the various housing solutions adopted in the emergency phase (Cas, hotels...), favouring a territorialist interpretation, taking into account the social and morphological features of this territory. The synthetic evaluation of the post-earthquake management tools that we are presenting here aims to investigate the territorial effects of the resulting housing chaos and to present the framework of inequalities and vulnerabilities raised by it

    Virtual biopsy in prostate cancer: can machine learning distinguish low and high aggressive tumors on MRI?

    In the last decades, MRI was proven a useful tool for the diagnosis and characterization of Prostate Cancer (PCa). In the literature, many studies focused on characterizing PCa aggressiveness, but a few have distinguished between low-aggressive (Gleason Grade Group (GG) =3) PCas based on biparametric MRI (bpMRI). In this study, 108 PCas were collected from two different centers and were divided into training, testing, and validation set. From Apparent Diffusion Coefficient (ADC) map and T2-Weighted Image (T2WI), we extracted texture features, both 3D and 2D, and we implemented three different methods of Feature Selection (FS): Minimum Redundance Maximum Relevance (MRMR), Affinity Propagation (AP), and Genetic Algorithm (GA). From the resulting subsets of predictors, we trained Support Vector Machine (SVM), Decision Tree, and Ensemble Learning classifiers on the training set, and we evaluated their prediction ability on the testing set. Then, for each FS method, we chose the best classifier, based on both training and testing performances, and we further assessed their generalization capability on the validation set. Between the three best models, a Decision Tree was trained using only two features extracted from the ADC map and selected by MRMR, achieving, on the validation set, an Area Under the ROC (AUC) equal to 81%, with sensitivity and specificity of 77% and 93%, respectively.Clinical Relevance- Our best model demonstrated to be able to distinguish low-aggressive from high-aggressive PCas with high accuracy. Potentially, this approach could help clinician to noninvasively distinguish between PCas that might need active treatment and those that could potentially benefit from active surveillance, avoiding biopsy-related complications

    Persistent warm Mediterranean surface waters during the Roman period

    Reconstruction of last millennia Sea Surface Temperature (SST) evolution is challenging due to the difficulty retrieving good resolution marine records and to the several uncertainties in the available proxy tools. In this regard, the Roman Period (1 CE to 500 CE) was particularly relevant in the socio-cultural development of the Mediterranean region while its climatic characteristics remain uncertain. Here we present a new SST reconstruction from the Sicily Channel based in Mg/Ca ratios measured on the planktonic foraminifer Globigerinoides ruber. This new record is framed in the context of other previously published Mediterranean SST records from the Alboran Sea, Minorca Basin and Aegean Sea and also compared to a north Hemisphere temperature reconstruction. The most solid image that emerges of this trans-Mediterranean comparison is the persistent regional occurrence of a distinct warm phase during the Roman Period. This record comparison consistently shows the Roman as the warmest period of the last 2 kyr, about 2 °C warmer than average values for the late centuries for the Sicily and Western Mediterranean regions. After the Roman Period a general cooling trend developed in the region with several minor oscillations. We hypothesis the potential link between this Roman Climatic Optimum and the expansion and subsequent decline of the Roman Empire

T-cell activation in vitro by pro-inflammatory cytokines may lead to functional Tissue Factor (TF) expression, indicating a possible contribution of immunity to thrombosis.     Receptor tyrosine kinase-dependent PI3K activation is an escape mechanism to vertical suppression of the EGFR/RAS/MAPK pathway in KRAS-mutated human colorectal cancer cell lines

    Abstract BACKGROUND: Previous studies showed that the combination of an anti-Epidermal growth factor (EGFR) and a MEK-inhibitor is able to prevent the onset of resistance to anti-EGFR monoclonal antibodies in KRAS-wild type colorectal cancer (CRC), while the same combination reverts anti-EGFR primary resistance in KRAS mutated CRC cell lines. However, rapid onset of resistance is a limit to combination therapies in KRAS mutated CRC. METHODS: We generated four different KRAS mutated CRC cell lines resistant to a combination of cetuximab (an anti-EGFR antibody) and refametinib (a selective MEK-inhibitor) after continuous exposure to increasing concentration of the drugs. We characterized these resistant cell lines by evaluating the expression and activation status of a panel of receptor tyrosine kinases (RTKs) and intracellular transducers by immunoblot and qRT-PCR. Oncomine comprehensive assay and microarray analysis were carried out to investigate new acquired mutations or transcriptomic adaptation, respectively, in the resistant cell lines. Immunofluorescence assay was used to show the localization of RTKs in resistant and parental clones. RESULTS: We found that PI3K-AKT pathway activation acts as an escape mechanism in cell lines with acquired resistance to combined inhibition of EGFR and MEK. AKT pathway activation is coupled to the activation of multiple RTKs such as HER2, HER3 and IGF1R, though its pharmacological inhibition is not sufficient to revert the resistant phenotype. PI3K pathway activation is mediated by autocrine loops and by heterodimerization of multiple receptors. CONCLUSIONS: PI3K activation plays a central role in the acquired resistance to the combination of anti-EGFR and MEK-inhibitor in KRAS mutated colorectal cancer cell lines. PI3K activation is cooperatively achieved through the activation of multiple RTKs such as HER2, HER3 and IGF1R

    A soil fungus confers plant resistance against a phytophagous insect by disrupting the symbiotic role of its gut microbiota

    Plants generate energy flows through natural food webs, driven by competition for resources among organisms, which are part of a complex network of multitrophic interactions. Here, we demonstrate that the interaction between tomato plants and a phytophagous insect is driven by a hidden interplay between their respective microbiotas. Tomato plants colonized by the soil fungus Trichoderma afroharzianum, a beneficial microorganism widely used in agriculture as a biocontrol agent, negatively affects the development and survival of the lepidopteran pest Spodoptera littoralis by altering the larval gut microbiota and its nutritional support to the host. Indeed, experiments aimed to restore the functional microbial community in the gut allow a complete rescue. Our results shed light on a novel role played by a soil microorganism in the modulation of plant-insect interaction, setting the stage for a more comprehensive analysis of the impact that biocontrol agents may have on ecological sustainability of agricultural systems

    Dual inhibition of TGFβ and AXL as a novel therapy for human colorectal adenocarcinoma with mesenchymal phenotype

    A subset of colorectal cancer (CRC) with a mesenchymal phenotype (CMS4) displays an aggressive disease, with an increased risk of recurrence after surgery, reduced survival, and resistance to standard treatments. It has been shown that the AXL and TGFβ signaling pathways are involved in epithelial-to-mesenchymal transition, migration, metastatic spread, and unresponsiveness to targeted therapies. However, the prognostic role of the combination of these biomarkers and the anti-tumor effect of AXL and TGFβ inhibition in CRC still has to be assessed. To evaluate the role of AXL and TGFβ as negative biomarker in CRC, we conducted an in-depth in silico analysis of CRC samples derived from the Gene Expression Omnibus. We found that AXL and TGFβ receptors are upregulated in CMS4 tumors and are correlated with an increased risk of recurrence after surgery in stage II/III CRC and a reduced overall survival. Moreover, we showed that AXL receptor is differently expressed in human CRC cell lines. Dual treatment with the TGFβ galunisertib and the AXL inhibitor, bemcentinib, significantly reduced colony formation and migration capabilities of tumor cells and displayed a strong anti-tumor activity in 3D spheroid cultures derived from patients with advanced CRC. Our work shows that AXL and TGFβ receptors identify a subgroup of CRC with a mesenchymal phenotype and correlate with poor prognosis. Dual inhibition of AXL and TGFβ could represent a novel therapeutic strategy for patients with this aggressive disease

    ‘See Me, Feel Me’: Prismatic Adaptation of an Alien Limb Ameliorates Spatial Neglect in a Patient Affected by Pathological Embodiment

    Pathological embodiment (E+) is a specific contralesional delusion of body ownership, observed following brain damage, in which patients embody someone else’s arm and its movements within their own body schema whenever the contralesional ‘alien’ arm is presented in a body-congruent position (i.e., 1st person perspective and aligned with the patient’s shoulder). This disorder is often associated with spatial neglect, a neurological syndrome in which patients are unaware of stimuli presented in the contralesional (often the left) space. Capitalizing on previous evidence demonstrating that prismatic adaptation of the ipsilesional arm to right-deviating prisms is effective in ameliorating neglect symptoms, here we investigated whether such amelioration also occurs in E+ patients with neglect when prismatic training is performed by the ‘alien’ embodied arm. Four left neglect patients (one with and three without pathological embodiment) underwent visuomotor prismatic training performed by an ‘alien’ arm. Specifically, while patients were wearing prismatic goggles shifting the visual field rightward, a co-experimenter’s left arm presented in a body-congruent perspective was repeatedly moved toward visual targets by another examiner. In a control condition, the co-experimenter’s arm was moved toward the targets from a body-incongruent position (i.e., 3rd person perspective). Neglect symptoms were assessed before and after training through paper-and-pencil tasks. In the E+ patient, neglect improved significantly more in 1st than in 3rd person perspective training, suggesting that prismatic adaptation of the ‘alien’ embodied arm is effective in modulating spatial representation. Conversely, for control E- patients (not embodying the ‘alien’ arm), we observed more limited improvements following training. These findings indicate that the ‘alien’ embodied arm is so deeply embedded in the patient body and motor schema that adaptation to prismatic lenses can affect multiple processing stages, from low level sensory-motor correspondences, to higher level body, motor and spatial maps, similarly as it occurs in normal subjects and neglect patients without pathological embodiment

    How future surgery will benefit from SARS-COV-2-related measures: a SPIGC survey conveying the perspective of Italian surgeons

    COVID-19 negatively affected surgical activity, but the potential benefits resulting from adopted measures remain unclear. The aim of this study was to evaluate the change in surgical activity and potential benefit from COVID-19 measures in perspective of Italian surgeons on behalf of SPIGC. A nationwide online survey on surgical practice before, during, and after COVID-19 pandemic was conducted in March-April 2022 (NCT:05323851). Effects of COVID-19 hospital-related measures on surgical patients' management and personal professional development across surgical specialties were explored. Data on demographics, pre-operative/peri-operative/post-operative management, and professional development were collected. Outcomes were matched with the corresponding volume. Four hundred and seventy-three respondents were included in final analysis across 14 surgical specialties. Since SARS-CoV-2 pandemic, application of telematic consultations (4.1% vs. 21.6%; p < 0.0001) and diagnostic evaluations (16.4% vs. 42.2%; p < 0.0001) increased. Elective surgical activities significantly reduced and surgeons opted more frequently for conservative management with a possible indication for elective (26.3% vs. 35.7%; p < 0.0001) or urgent (20.4% vs. 38.5%; p < 0.0001) surgery. All new COVID-related measures are perceived to be maintained in the future. Surgeons' personal education online increased from 12.6% (pre-COVID) to 86.6% (post-COVID; p < 0.0001). Online educational activities are considered a beneficial effect from COVID pandemic (56.4%). COVID-19 had a great impact on surgical specialties, with significant reduction of operation volume. However, some forced changes turned out to be benefits. Isolation measures pushed the use of telemedicine and telemetric devices for outpatient practice and favored communication for educational purposes and surgeon-patient/family communication. From the Italian surgeons' perspective, COVID-related measures will continue to influence future surgical clinical practice
